Matthews shared an opening stand of 128 with Realeanna Grimmond, who played an important supporting role with a composed 53 from 70 balls.
Grimmond initially provided the greater aggression, taking medium-pacer Tendai Makushka for a boundary and a six as she moved into the 30s.
Matthews soon joined the attack.
She drove off-spinner Nyasha Gwanzura for three boundaries in the 15th over, taking herself to 42 while pushing the West Indies to 92 without loss.
The openers then carried their team beyond 100, with both reaching half-centuries before Zimbabwe finally found a breakthrough.
Spinner Beloved Biza ended the partnership when Grimmond edged behind for 53, an innings containing seven fours and a six.
Taylor falls as Matthews takes control
Stafanie Taylor lasted only two runs before Francisca Chipare bowled her, leaving the West Indies at 139 for two.
That setback barely interrupted Matthews.
She added 48 with Shemaine Campbelle, who contributed 13, steadily taking the West Indies toward a formidable total.
Then came the milestone.
Matthews reached her 13th ODI century by driving Gwanzura through cover for two runs before raising her bat toward the dressing room.
But the hundred was only the beginning.
A record innings gathers pace
Gwanzura soon struck again, trapping Campbelle lbw, but Matthews shifted through the gears alongside Chinelle Henry.
The pair tore through 71 runs in fewer than 10 overs, pushing the West Indies to 258 for four in the 46th over.
With Matthews still at the crease, 300 appeared within reach.
Instead, Zimbabwe finally stopped the assault when Makushka bowled Matthews with the first delivery of the final over.
By then, however, the damage was immense.
Matthews had faced 156 balls and struck 26 fours and two sixes, surpassing her previous West Indies record ODI score of 159 not out against Ireland in July.
Chipare was Zimbabwe's most successful bowler, finishing with 2-57 from her 10 overs.
Zimbabwe threaten before Fletcher changes the game
Zimbabwe's chase initially showed signs of resistance.
Sharne Mayers made 21 and Kelis Ndhlovu 15 before Modester Mupachikwa anchored the response with a determined half-century.
Mupachikwa and Chipo Mugeri-Tiripano then put Zimbabwe in a promising position, moving the hosts to 100 for two in the 22nd over.
The West Indies needed a breakthrough, and Afy Fletcher provided it.
She ended the 49-run partnership by having Mugeri-Tiripano caught for 26, and Zimbabwe's innings began to lose its shape.
Fletcher then removed Beloved Biza for 11, while captain Josephine Nkomo was run out for 10.
Zimbabwe had fallen to 152 for five by the end of the 34th over.
Five wickets disappear for 48
From there, the chase unraveled quickly.
Zimbabwe lost its final five wickets for only 48 runs, with Mupachikwa left to fight almost single-handedly.
She eventually fell for an admirable 86 from 106 balls, including seven fours and a six, as the hosts were bowled out for 200 in the 44th over.
Aaliyah Alleyne led the West Indies attack with 3-44, while Ashmini Munisar, Fletcher and Jannillea Glasgow claimed two wickets each.
The result gave the West Indies a 1-0 lead and ensured that Matthews' record-setting performance was rewarded with a comprehensive victory.
More importantly, the visitors now have the opportunity to close out the three-match series with another strong performance after announcing their intentions in emphatic fashion in the opener.
